Septin 8 interacts with septin 4 and septin 5; these proteins are concomitantly expressed in platelets (6). Sept8 is localized surrounding the alpha-granules and is implicated in platelet granular secretion (7). Sept8 is also required for distribution of sept5 near the vesicles in the cytoplasm of cells. In the absence of sept8, sept5 was found to be diffused throughout the cytoplasm whereas in the presence of sept8, these two proteins were colocalized to the vesicular location in the cytoplasm (8). Sept8 has a long N-terminal extension which is rich in proline residues and a C-terminal coiled-coil domain (9). Septin 8 is located on mouse chromosome 11B1.3. It is expressed in brain, heart, platelets and other tissues with high expression in aorta. Septin 8 is approximately a 51.6kDa protein (430 amino acids).

The Sept8 antibodies were generated using peptide corresponding to mouse Sept8 protein. Sept8 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Inc. will provide limited quantities of antigenic blocking protein for competition assays involving Sept8 antibodies. Antibodies to Sept8 (Sept-801AP) will label ~56kDa protein in Western blot positive control for Sept8 (PC-Sept8) and several other tissues.  FabGennix Inc. will conjugate this and other antibodies from its catalog to either secondary enzymes (alk-Pase or HRP) or fluorescent probes at a nominal cost upon request.  FabGennix also provides custom antibody production services for researchers that are looking for high affinity mono and polyclonal antibodies in various species. We specialize in making application specific antibodies that are useful in IHC, confocal and other applications where native antigen is detected. Septin 8 (Sept8) belongs to the septin family of GTPases. GTPases are central regulators in signal transduction pathways in every aspect of cell biology (1). Septins bind to and hydrolyze GTP and most of these proteins polymerize to form filaments which may form cytoskeletal system that organizes higher-order structures (2). Septins were first discovered in yeast Saccharomcyes Cerevisiae, they are also found in animals and fungi but not in protozoa or plants (3). Septins are implicated in cytokinesis, exocytosis, tumorigenesis, apoptosis, synaptogenesis, neurodegeneration, and in functions involving changes in cell shape and in the specialized regions of the cell cortex (4). Humans have twelve septin genes, some of which generate multiple polypeptides either by alternative splicing or alternative translation start sites (2). The function of individual protein in septin family usually depends on its assembly into oligomeric complexes and highly ordered polymers (5).
