Septins are conserved GTP binding proteins important for cytokinesis, membrane compartmentalization and exocytosis.  Septins belong to a ever growing family of proteins (Septin 1-14) that are rapidly exchange subunits and rearrange with in the cytoplasm and this mechanism  is conserved form yeast to mammals.  There are many subtypes of septins GTPases are central regulators in signal transduction pathways in every aspect of cell biology (1). Septins bind to and hydrolyze GTP and most of these proteins polymerize to form filaments which may form cytoskeletal system that organizes higher-order structures (2). Septins were first discovered in yeast Saccharomcyes Cerevisiae, they are also found in animals and fungi but not in protozoa or plants (3). Septins are implicated in cytokinesis, exocytosis, tumorigenesis, apoptosis, synaptogenesis, neurodegeneration, and in functions involving changes in cell shape and in the specialized regions of the cell cortex (4). Humans have twelve septin genes, some of which generate multiple polypeptides either by alternative splicing or alternative translation start sites (2). The function of individual protein in septin family usually depends on its assembly into oligomeric complexes and highly ordered polymers (5).

Septin 6 has been shown to play an important role during exocytosis. Members of Septin 6 family interacts with Septin 2, along with Septin 6. More specifically, it interacts with syntaxin and synaptophysin in the process of membrane fusion during exocytosis (6). Septin 5 also interacts with Septin 11; this interaction requires the GTP-binding domain and the C-terminal extension (7). Septin 5 is also implicated in the development of autosomal-recessive juvenile Parkinsonism (AR-JP). It has been reported that inhibition of dopamine secretion by this protein may contribute to the development of AR-JP (8). Septin 5 is located on human chromosome 22q11.21. It is highly expressed in the CNS, heart and platelets. Septin 6 is approximately a 52kDa protein (429 amino acids).  Septin 6along with hnRNP inhibits the Hepatitis C virus replication (6).

The Sept6 antibodies were generated using peptide corresponding to human Sept6 protein. Sept5 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Immunogen:            Synthetic peptide corresponding to unique epitope on Sept6.  The peptide sequence was selected from C-terminal, amino acids 387-411.  These peptides were covalently modified post-synthetically to achieve desired antigenicity.

Concentration:         Sept-601AP:  IgG concentration 0.54-0.59 mg/ml in antibody stabilization buffer.

Applications:           Antibody Sept-601AP is ideal for WB and ELISA applications, other applications have not been tested. The species cross reactivity for these antibodies have not been examined fully.  The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions for specific assay.  WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d.  ELISA <1:10,000.  Application of this antibody in protocols not listed here does not necessarily exclude its use in such procedures.  Investigators using this antibody in applications not listed here can ask for a free sample of this antibody.

Reactivity:               This antibody detects a single band of approximately 52kDa in PC-Sept6 samples.
