The National Institutes of Health Mammalian Gene Collection (MGC) Program is a multi-institutional effort to identify and sequence a cDNA clone containing a complete ORF for each human and mouse gene. ESTs were generated from libraries enriched for full-length cDNAs and analyzed to identify candidate full-ORF clones, which then were sequenced to high accuracy. The MGC has currently sequenced and verified the full ORF for a non--redundant set of >9,000 human and >6,000 mouse genes. Candidate full-ORF clones for an additional 7,800 human and 3,500 mouse genes also have been identified. All MGC sequences and clones are available without restriction through public databases and clone distribution networks (see http:mgc.nci.nih.gov). the FLJ23499 contains a DUF domain present in many other proteins, the function of this domain is not known.
Certain loci on human chromosome 11q13-q22 are frequently amplified in various types of cancers. One of such loci is PP1R1B-ERBB2-GRB7 which is greatly amplified in gastric and breast cancer. Search of other genes that reside close to this recombination hot spot or “fragile loci” led to discovery of several genes, one of these genes is a new theoretical protein FLJ23499 is localized on chromosome 11 at cytogenetic location of 11q13-q22.   The detailed analyses of the amplicon structure in 330 breast tumors samples revealed a 280 kb common region of amplification that contains 10 genes that include 8 known genes. A highly significant association between amplification and expression levels was observed for 6 genes including ERBB2 and 2 uncharacterized hypothetical proteins, MGC9753 and MGC14832 in breast carcinoma and human gastric cancer samples (1, 2). FLJ23499 is located on human chromosome 11q13-q22 and is in the amplicon cluster consist of PPP1R1B-STARD3-TCAP-PNMT-MGC9753-ERBB2-MGC14832-GRB7 with in the 120 kb region locus on human chromosome 17q12. The FLF23499 protein is a soluble protein with no evidence of any TMD. The human FLJ23499 protein is a 150 amino acid protein.
The FLJ23499-selective (C11Orf1) antibodies were generated in rabbits against synthetic N, mid-region and C-terminal peptides that are unique to FLJ23499 protein. The anti-FLJ-23499 antibodies were affinity purified on respective iimobilized antigen affinity matrices and purified antibodies were stabilized for long-term storage. Limited quantities of antigenic blocking peptides for multiple FLJ23499 antibodies are also available. FabGennix will conjugate FLJH23499 antibodies to fluorophores, secondary enzymes or biotin at a nominal cost. Some of these conjugates are already made are in stock.
