Biomarkers for identification of certain diseases play an important role in management of various pathological conditions of diseases. This involves diagnosing disease, predicting disease progression and monitoring the efficacy of treatment modalities. The disease specific markers are still scarce, despite the efforts to identify these markers is picked up. PTK7 is one of such potential biomarker for T-cell acute lymphoblastic leukemia (1). Receptor tyrosine kinases are trans-membrane signaling proteins that transduce extracellular signals to the interior of cells. Aberrations in these signaling molecules results in development and progression of many types of cancers.
Colon carcinoma kinase 4 (CCK4), also known a s protein tyrosine kinase-7 (PTK7) also is a catalytically inactive receptor tyrosine kinase (RTK) that is up-regulated in many types of cancer including colon, lung, gastric and acute myeloid leukemia. Since CCK4 is defective in several motifs important of kinase activity, it is suggested that it might have a role in amplification or modulation of signal for tumor progression (2). The levels of CCK4/PTK7 are reported higher in CD34+ hematopoietic progenitors and in AML (3), in contrast, metastatic melanomas had low or no expression. CCR4/PKT7 is a novel regulator of non-canonical WNT or planar cell polarity signaling for formation, migration and invasion of endothelia and angiogenesis in HUVEC cells (4). CCK4 is an approximately 130kDa (1070aa) protein, the gene is located on chromosome 6p21.1 deletions of this chromosome are frequently reported in a number of cancers. CCK4 has an extracellular domain with 7 immunoglobulin-like loops, a trans-membrane domain, and a catalytically inactive tyrosine kinase domain.
The CCK4 antibodies were generated using purified protein. The anti-CCK4 antibodies were pre- absorbed to remove non-specific antibodies and then affinity purified on immobilized CCK4 affinity column. The affinity purified immunoglobulins are stabilized in antibody stabilization buffer for long-term storage. FabGennix Int. Immunogen: Purified Protein tyrosine kinase 7 protein was emulsified in complete adjuvant and used to immunize rabbits. The anti-serum was pre-cleared for tag protein and then affinity purified on immobilized PTK7 affinity chromatography. The purified immunoglobulins are stabilized in patented antibody stabilization buffer for long-term storage.
Concentration: CCK4-401AP and CCK4-FITC: IgG concentration 0.57-0.63 mg/ml in antibody stabilization buffer.
Applications: Antibodies PTK7-101AP is ideal for WB and ELISA applications, other applications have not been tested. The species cross reactivity for these antibodies have not been examined fully. The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions for specific assay. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d. ELISA <1:10,000. Application of this antibody in protocols not listed here does not necessarily exclude its use in such procedures. Reactivity: This antibody detects a single band of approximately 135kDa in PC-CCR4 samples.

Form/Storage: The antiserum is supplied in antibody stabilization buffer. The affinity-purified antibodies are isolated on immobilized antigen-affinity column and supplied as stabilized product. Store at -20oC for long-term storage. FabGennix Inc. does not recommend storage of very dilute antibody solutions unless they are prepared in specially formulated multi use antibody dilution buffer (Cat # DiluOBuffer). Working solutions of antibodies in DiluOBuffer should be filtered through 0.45um filter after every use for long-term storage.
