Runt-related Transcription factor1 (Runx1) belongs to the family of Runt-related  to a transcription factors comprising of 3  subtypes (Runx1-3).  Regulation of cell proliferation by RUNX proteins represents an evolutionarily conserved activity.  All vertibrates ave three Runx genes and all three genes have P1 and P2 promotors. The Runx3 P1 and P2 promoter regions showed that a small (around 600 bp) P1 element could direct cell-type-specific expression with fidelity (1).   Runx1- transcription factor is an essential regulator of hematopoiesis and has multiple isofroms arising from differential splicing and utilization of two promotors. At least three variants have been characterized for Runx1 (Runx1a, b and c).  all Runx transcription factors (Runx1, Run2 and Runx3) bind to the same DNA motif, hence their  temporal and spatial expression is during development is tightly regulated.  The c isoform is transcribed from the use of distal promotor that resulted in a transcript that have 32 amino acids unique to this isoform (2)1).  The Runx1b has 5 unique N-terminal amino acids and originated form the use of proximal promotor P2.  Runx1 is a powerful regulator of hematopoiesis.   Runx1 transcription factor is highly expressed at a CD4/CD8-double-negative (DN) stage of thymocyte development but is down-regulated when cells proceed to the double-positive (DP) stage (3).

Runx1 function can be regulated by post-translational modifications (arginine methylation by PRMT1) and protein–protein interactions.  The PRMT1 methylation of R206 and R210 on Runx1 abrogated its association with corepressor SIN3A (4).  Runx1 mutation has been  seen in patients with familial platelet disorders, thrombocytopenia.  RUNX1 also directly regulates G1 to S cell cycle progression of the hematopoietic cells, which is accounted for by the phosphorylation of  S303 by cdks lead to ubiquitin-mediated degradation during G2/M phase (5).  Runx1 is a 58kDa protein (480aa).  There are several isoforms that are expressed in hematopoietic cells.

Runx1- selective antibodies were generated against a peptide taken form N-terminal region spanning 1-75 amino acids. The 23mer peptide was conserved in Runx1 isoform 1, 2 and 3 of human Runx1.  The antibodies do not cross react to onther Runx subtypes. The anti-Runx1-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions.  The Runx1antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage.  FabGennix Int. Immunogen:                     The 23mer synthetic peptide taken from human Runx1 protein  from within the region between aa 1-75.  The peptide was conserved in several other species.  The selected peptide was post-synthetically modified to achieve highest antigenicity before coupling to carrier protein using hetero bifunctional cross linker for immunogen preparation.                  

Conc.:        Affinity purified Runx1-101AP and Runx1-FITC antibodies were stabilized at a concentration 0.51-0.53mg/ml.  .

Appl.:        Runx1-101AP antibody was tested in ELISA and western blotting applications at 1:500 dilution using PC-Runx1 samples.  Antibody dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions.  Application of this antibody in other protocols has not yet tested. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d.  Investigators using this antibody in protocols other than listed above can request a complimentary sample of this antibody. N.D not necessarily means the antibody is not suitable for that application, it simply means we have not yet characterized the antibody for that application.

Reactivity:  The antibody labels a strong band of Runx1of 58kDa in PC-Runx1samples and in several cell lines. The antibody also labels diffuse bands of around 90 kda, the identity of these band is not known but may represent Runx 1 splice variants.
