Deleted in Colorectal Carcinoma protein (DCC) and Neogenin are receptors for Netrin 1 in developing brain. DCC is a tumor suppressor gene as well as a conditional oncogene (1). When DCC is bound to its ligand, netrin, it acts as oncogene and activates the pathways that promote cell survival and nuerite growth down-stream to ligand concentration. Halo DCC promotes apoptotic pathways (2,3). When DCC is not bound to netrin 1 it can initiate apoptosis without the formation of an apoptosome or cytochrome c release (4). DCC has recently been found to suppress metastasis in vitro on cells. DCC trans-membrane protein is a member of the immunoglobulin superfamily of cell adhesion molecules, and mediates axon guidance of neuronal growth cones towards sources of netrin 1 ligand.
DCC is a 1447 aa protein resides on chromosome 18q21. DCC is expressed in low abundance ubiquteously and at higher levels in many areas of the brain, particularly in dopaminergic neurons. It is found in axons of the central and peripheral nervous system and in differentiated cell types of the intestine. Higher levels of DCC are found in neonatal tissues and its expression is significantly reduced in adult hood (3). The DCC is expressed on developing spinal cord neurons to promote axonal and nuerite growth in response to netrin. DCC and neogenin, two of the netrin-1 receptors, have recently been shown to have sites for tyrosine phosphorylation (at Y1420 on DCC) and are likely interacting with Src family kinases in regulating responses to netrin-1(5). The DCC undergoes proteolytic degradation to yield a 120 kDa fragment in apoptotic cells (6).
The DCC-selective antibodies were generated against antigenic peptide taken from human DCC protein corresponding near to carboxy terminal end of DCC protein. DCC- antibodies are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Inc., will also provide limited quantities of antigenic blocking protein for DCC-101AP antibody. Antibodies to DCC (DCCR -101AP) labels ~180kDa protein and ~ 120 kDa in PC-DCC samples.
