TNF receptor-associated factor 6 (TRAF6), located at human chromosome 11p12, is a member of the TRAF protein family. TRAF proteins mediate signal transduction from members of tumor necrosis factor (TNF) receptor superfamily and Toll/IL-1 family, and also from receptors such as CD40, TNFSF11/RANCE and interleukin-1 (IL-1) (1). More specifically, TRAF6 is an intracellular signaling adaptor molecule for TNF receptor superfamily members such as CD40 and Toll like receptors (TLRs) (2). It acts as a signal transducer for IL-1 and activates nuclear factor-kappa B (NF-kappa B) (3). TRAF6 plays a critical role in T-cell dependent (TD) and T-cell independent (TI) humoral immune responses and in the maturation of B cells (1). TRAF6 is also known to regulate cell fate decisions by inducing either apoptotic or anti-apoptotic pathways (4). In addition to TNFR and IL-1/TLR superfamily signaling, TRAF6 is crucial in the development of several tissues including lymph nodes, mammary glands, skin and the central nervous system (5).

Deficiency of TRAF6 protein in mice resulted in the reduction of mature B cells in bone marrow and spleen and these mice failed to develop mature dendritic cells (DCs). These mice also exhibited lower levels of serum IgM, IgG2b and produced defective antigen-specific IgM in response to TI antigens (1). In response to microbial components, TRAF-6 deficient DCs failed to produce inflammatory cytokines (6). Therefore, TRAF6 is important for adaptive and innate immunity and also in bone homeostasis. TRAF-6 is expressed in normal and diseased tissue. It is approximately a 62.6kDa protein (522 amino acids).

The TRAF6-selective antibodies were generated against purified Glutamate receptor 1 (TRAF6) protein. TRAF6-antibodies are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Inc., will also provide limited quantities of antigenic blocking protein for TRAF6 -101AP antibody. Antibodies to TRAF6 (TRAF6 -601AP) will label ~109kDa protein in Western blot positive control samples for TRAF6 and several other tissues. FabGennix Inc. will also conjugate antibodies with secondary enzymes (alk-Pase or HRP) or fluorescent probes upon request at nominal cost.
