Endocannabinoids (eCBs) are endogenous lipid mediators capable of binding to and functionally activating G-protein coupled cannabinoid receptors (CB1 and CB2) to modulate synaptic function. Both CB1 and CB2 receptors belong to the superfamily of 7TMD containing G protein-coupled receptors (GPCRs) and act mainly through inhibitory G proteins (Gi/o). Activation of CB1/CB2 led to a decrease in cellular cAMP levels. Activation of CB1/CB2 can activate post receptor signaling pathways mediated by different kinases activating nuclear factor-kappaB (NF-kappaB), nuclear factor of activated T cells (NFAT) and activator protein-1 (AP-1) DNA binding. Binding of agonist to CB1/CB2 receptors led to epigenetic regulation of several genes. CB1/Cb32 receptors also exhibited anti-nociceptive action when cannabinoids are injected directly into the hind paw skin of uninjured rats..
The cannabinoid receptors CB1/CB2 are expressed in CNS and in peripheral tissues including cells of hematopoietic origin, expressed in skin in suprabasal and hair follicles, in brain by neurons and glial cells and by osteoblasts, osteocytesm osteoclasts. Both receptors are localized in nerve fibers of keratinocytes, dorsal root ganglion tissue, and in other peripheral tissues. CB2 is expressed predominantly outside the CNS and is markedly up-regulated after spinal nerve ligation. The cannabinoid delta-9-tetrahydrocannabinol (THC) binds to CB1/CB2 receptors and produce psychodelusional effects. The amino acid sequence homology between CB1 and CB2 is unusually low for GPCR that bind the same neurotransmitter/neuromodulator ligand. The role of CB2 receptor in CNS in addiction and major depression is ambiguous and controversial (1). The cannabinoid system is immunomodulatory and has been targeted as a treatment for the central nervous system (CNS) autoimmune disease multiple sclerosis (2). CB2 receptor is located on chromosome and has apparent MW of 42kDa comprising of 347aa.
The CB2R antibodies were generated using peptide corresponding to mouse CB2R protein. Immunogen: Synthetic peptide unique to CB2 receptor and common in both CB2L and CB2S type receptors. The epeptide reside between amino acids 300-347. The peptide was covalently modified to obtained desired antigenicity before conjugating to a carrier protein.
Concentration: CB2R-201AP and CB2R-FITC: IgG concentration 0.61-0.63mg/ml in antibody stabilization buffer.
Applications: Antibody CB2R-201AP is ideal for WB and ELISA applications, other applications have not been tested. The species cross reactivity for this antibody has not been fully elucidated. The dilution for this antibody is for reference purposes only, investigators are expected to determine the optimal conditions for a specific assay. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d. ELISA <1:10,000. Application of this antibody in protocols not listed here does not necessarily exclude its use in such procedures.
