Cadherin 74A/B

Accession Number and Alternate nomenclature:  NP_684973; CG64450PA and CG6445-PB.

Cad74A is one of 17 genes in the Drosophila genome encoding putative cadherin proteins and has 13 cadherin domains, a N-terminal signal peptide, a transmembrane domain, and a short uncharacterized cytoplasmic domain (1).  Cadherin are glycoproteins involved in Ca-mediated cell-cell adhesion and paly a role  in cell fate signaling, proliferation, differentiation, and migration. There are several members in Cadherin family, including E-, N-, P-, VE-, CNR-, and desmocollin and desmoglein. FAT Cadherin74A (Cad74), is a non-classical cadherin, function as a putative adhesion gene that bridges epithelial patterning and morphogenesis in the follicle cells. Cad74 is expressed in the follicle cells that contact the oocyte, including the border cells and most of the columnar follicular cells. The expression of Cad74A is repressed in dorsolateral patches of the follicle cells that participate in the formation of respiratory appendages.  Complete removal of Cad74A form the follicular cells leads to defects in the dorsal appendages formation.  Cad74A serves as adhesive machinery that enables robust dorsal appendages formation and also provides a link between follicle patterning cells and eggshell morphogenesis (2).

So far the functional characterization of CXad74A/B  is not fully understood but it is proposed that Cad74Ais a component in the follicular morphogenetic machinery that translates two dimentional patterns in to three dimentional egg patterns. .  Cad74A mediates calcium-dependent homotypic cell-cell adhesion in cultured cells and is located sub-apically and apically in vivo. Cad74A is also expressed in early embryo, in the nerectodrem, the developing drosophila larval brain and in eye imaginal disc.  Cad74A/b are 1820 aa acid long protein with ca binding sites. 

The D. melanogaster CadherinA/B-selective antibodies were generated against unique antigenic sequence taken form the C-terminal region with in the amino acids (1669-1793 and 1775-1793.  These peptide sequences were conserved in both Cad74A/B s of fruit fly, but was unique to only Cad74 protein and was not present in other proteins.  The Cadh74-antibodies were affinity purified over immobilized antigen based chromatography, and the affinity purified immunoglobulins are stabilized in antibody stabilization buffer.  FabGennix Int. Immunogen:    Synthetic peptide taken form the Cad74A/B protein corresponding to peptide seq (clk kkl krh skr slqa slr) and (ldt qvf trp dms ldf tat ssc) form within the C-terminal region, the peptides were post-synthetically modified and conjugated to carrier protein to achieve desired antigenicity.  The Shade peptides sequences are conserved various other species of fruit flies.

Concentration:         SCad74-101AP and Cad74-112AP:  IgG concentration 0.55-0.68mg/ml in antibody stabilization buffer.

Applications:           Antibody Cad-101AP and Cad74-112AP are ideal for WB and IHC applications, other applications have not been tested.  This antibody does not cross react to other fly proteins. The species cross reactivity for this antibody is not examined fully.  The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions for specific assay.  WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d.  ELISA <1:10,000; n.d does not necessary means antibody will not work in that applications.  Investigators using this antibody in applications not listed here can request for a complimentary sample. 

Reactivity:               This antibody detects a diffuse band of approximately ~225kDa PC-cad74A/B samples.  n.d does not necessarily means antibody is not suitable for that application, it simply indicate it has not been tried for that application. Investigators using this antibody in applications not listed here can ask for a free complimentary sample of this antibody.

Protocols:                Standard protocol for various applications (WB; IMM and IHC) of this antibody can be requested by calling Technical support line, basic information on this antibody is provided with the product specification sheet, however, FabGennix Inc.  strongly recommends investigators to optimize conditions for use of this antibody in their laboratories.  

Form/Storage:         The affinity-purified antibodies are supplied as stabilized product.  Store at -20^oC for long-term storage.  FabGennix Inc. does not recommend storage of very dilute antibody solutions unless they are prepared in specially formulated multi use antibody dilution buffer (Cat # DiluOBuffer).  Working solutions of antibodies in DiluOBuffer should be filtered through 0.45u filter after every use for long-term storage.
