Pax transactivation domain-interacting protein (PTIP) is a ubiquitously expressed, nuclear protein that is part of a histone H3K4 methyltransferase complex and is essential for embryonic development. PTIP is an essential element of the cell proliferation machinery by functioning in the DNA repair pathway. Methylation of H3K4 is an epigenetic marker often found in many critical developmental regulatory genes in embryonic stem cells and together with H3K27 constitute a bivalent epigenetic signature. PTIP function is required for constitutive methylation of H3K4 during in vitro propagation of pluripotent embryonic stem cells (1). PTIP is a large nuclear protein with multiple BRCT domains that are identified by their interaction with transcription factor PAX and Smad (2). PTIP also plays a critical role in adipogensis by regulating PPARgamma and C/EBPalfa expression (3).
PTIP1 contains six BRCT (BRCA1 carboxy terminal) implicating its role in DNA damage response. Exposure of cells to ionizing radiation, PTIP forms a nuclear foci and physically interacts with DNA repair factor 53BP1 (4). PTIP also promotes Ig switch region transcription initiation at a stage preceding pre mRNA splicing to promote initiating Igh switch transcripts expression (5). PTIP is a 125 kDa (1025 aa) nuclear protein expressed in many developing embryonic pluripotent cells.
The PTIP antibodies were generated using peptide corresponding to C-terminal domain of the human PTIP gene. The PTIP antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Antibodies to PTIP (PTIP-101AP) will label ~130kDa protein in Western blot positive control for PTIP (PC-PTIP).
