Activating Transcription Factor 2
Activating Transcription Factor 2 (ATF-2) is a member of transcription factirs that have characteristic “bZip” motifs thereby regulating gene expression. ATF2 has been shown to regulate gne expression in cellular response to stress (1). Heterodimer formation between members of the bZip group is common and is believed to add diversity to the cis-acting elements at which binding of the dimers is directed. Specifically, ATF-2 may dimerize with c-Jun, as occurs in response to E1a, and in so doing shift the binding preference of c-Jun toward ATF/CRE sites (2-4). Deletion analysis has indicated that the N-terminal region of ATF-2 containing threonine at residues 69 and 71 are essential for this purpose. These threonine residues are phosphorylated by JNK/SAPK for transcriptional activation.
The ATF2 is also substrate for phosphorylation by vaccinia related kinase 1 (VRK1) in human kinome (5). The VRK1 phosphorylates ATF2 that regulates gene expression by forming dimers with other transcription factors recognizing CRE or AP-1 sequences implicated in cellular responses to stress (5). ATF2 phosphorylation was decreased by antidepressants and increased at the aftermath of prolonged stress, specifically in the frontal cortex (6).
The Phopsho-ATF2-selective antibodies were generated using a synthetic peptide containing phosphorylated Thr-71 of human ATF2. The mouse monoclonals are of isosubtype IgG1 stablized in antobody stabilization buffer containing 0.02% sodium azide as preserivative. The ascites are affinity purified on a protein G affinity matrix. FabGennix Int.
