Serine proteases are well known as enzymes involved in digestion of dietary proteins, blood coagulation, and homeostasis. A new role of serine proteases as signal molecule acting via protease-activated receptors (PARs) was recently uncovered. Important effects of PAR activation on leukocyte motility, cytokine production, adhesion molecule expression, and a variety of other physiological or pathophysiological functions have been described in vitro and in vivo. PAR activation is also involved in a variety of diseases conditions including neuroinflammatory and neurodegenerative process of skin, joints, and airway inflammation. Additionally Par activation is involved in innate and adaptive immunity (1). PAR receptors belong to the 7TMD family members and are expressed by 4 distinct genes (PAR1-4).
PAR receptors and purinergic receptors (P2RY12 ) play important role in homostasis and thrombosis. Par3knockout mice showed nearly complete protection against carotid artery thrombosis caused by low FeCl(3) injury. Par3 and Par4 heterozygous mouse platelets showed partially decreased responses to thrombin, resembling those in PAR1 antagonist-treated human platelets (2). PAR3 is a thrombin receptor that is expressed in mouse megakaryocytes and is responsible for normal thrombin responses in mouse platelets (3).
The PAR 3 selective antibodies were generated using a peptide selected form region between amino acids 31-55. The PAR3 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer for long-term storage. FabGennix Int. Inc. will provide limited quantities of antigenic blocking peptides for competition assays involving PAR3 antibody. Antibodies to PAR3 label ~48kDa protein in Western blot positive control and several other tissues. FabGennix Inc. will conjugate this and other antibodies from its catalog to either secondary enzymes (alk-Pase or HRP) or fluorescent probes at a nominal cost upon request. FabGennix also provides custom antibody production services for researchers that are looking for high affinity mono and polylconal antibodies in various species. We specialize in making application specific antibodies that are useful in IHC, confocal and other applications where native antigen is detected. Immunogen: Synthetic peptide from amino acids 31-355 corresponding to rat PAR2 protein from the following sequence (KPTLTIKSFNGGPQNTFEEFPLSDIE ), was selected from the unique region of the mouse PAR3 protein, peptide was post-synthetically modified to achieve highest antigenicity before used for coupling to KLH using heterobifunctional cross linker for immunogen preparation.
Concentration: PAR3-301AP Ig concentration 0.51-0.54 mg/ml in antibody stabilization buffer.
Applications: Antibody PAR3-301AP is ideal for IMM/WB applications for detection of endogenous PAR3 protein in PC-PAR3 samples. The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions. Western blotting: > 1:250; IMM: Immunprecipitation1:200 recommended; IHC = nd. Investigators who want to use this antibody in applications not listed here can ask for a complimentary sample of PAR3 antibodies. We will be happy to provide this antibody form multiple rabbits.
Reactivity: This antibody detects a band of approximately 48kDa PAR3 protein in PC-PAR3 samples. The antibody also labels a 58kDa bands in PC-PAR3 samples, probably represents either an uncharacterized variant of PAR3 or a non-specific protein. Further experiments are needed to address these possibilities.
