Phosphodiesterase Type 4A5 antibody

The PDE4 family of cAMP-specific phosphodiesterases represents an important therapeutic and pharmacological target for a variety of therpapeutic interventions including their anti-inflammatory and anti-depressant effects.  Four distinct genes (PDE4A, PDE4B, PDE4C and PDE4D), encode large family of PDE4 isoforms [1, 2).  Almost all of these isoforms have a common core made up of central catalytic domain and C-terminal tail.  The differential RNA splicing and use of alternate initiation codons also generates the elongated N-terminal ends “long-forms” PDE4 proteins.  The long form PDE variants are characterized by upstream conserved regions (UCR1 and UCR2).    The PDE4A5 is a rat/mouse homologue of human PDE4A4 long-form PDE4A variant.  The PDE4A5 has the highest sensitivity to rolipram inhibition (3nM) that is at least 100-fold less than for other PDE4 family members in U937 cells (3).
PDE4A5-selective antibody was generated against unique N-terminal sequence, the peptide was post-synthetically modified to achieve maximum antigeneicty and provide proper linkers for covalent coupling to carrier protien (blue carrier protien).  The anti-PDE4A5 antibodies does not cross-react with other PDE4A proteins including A4, A1 and Ax.  Antibody PD4-150 is also available in affinity-purified form (Cat # PD4-151AP) and as HRP conjugate (PD4-152HRP) for western blotting and immunnohistochemical localization experiments.  FabGennix Inc. provides PDE family selective, family subtype-selective and subtype-variant selective antibodies for detailed analyses of cAMP signaling pathways.  The peptide was conjugated with BCP for the production of antibodies in rabbits.   Antibody PD4-150P and PD4-151AP labels PDE4A5 protein in PC-PD4A5 samples, rat hippocamplal extrcts and in SHSy-5Y cells.  The PDE4A5 variant specific antibody does not cross react with PDE4A8 or other PDE4A proteins including PDE4A4, PDE4A1 and PDE4Ax or with other PDE family members.  FabGennix Inc. carries Western blot positive controls for PDE4A5 in ready-to-use buffers for easy identification and quantification of PDE4A4 protein in samples.   FabGennix Inc. also offers services to tag a fluorophore or make custom affinity matrices using these antibodies.  FabGennix Inc. also offers a kit for rapid isolation of testicular germ cells (stem cells) for short-term primary cultures using fluorescence-activated cell sorting (FACS) technique.
