Sestrin 1(SESN1), located on rat chromosome 20q13, belongs to the sestrin family of proteins. These proteins are cystein sulfinyl reductases and they modulate peroxide signaling and antioxidant defense (1). This family of proteins is only present in multicellular organisms ranging from nematodes to mammals. These proteins selectively reduce or repair hyperoxidized forms of typical 2-Cys peroxiredoxins within eukaryotes (2). Peroxiredoxins are the enzymes that metabolize peroxides. Expression of these proteins is regulated by p53, a tumor suppressor protein (3). Sestrin 1 is implicated in the inhibition of cell growth. It has been reported that Sestrin 1 activates AMP-responsive protein kinase (AMPK) and targets it to phosphorylate TSC2, a tumor suppressor. Phosphorylation of TSC2 stimulates its GAP activity which inhibits the activity of mTOR, a positive regulator of cell growth, thus leading to inhibition of cell proliferation and growth (4).
Inhibition of mRNAs from sestrin 1 and 3 genes has been shown to substantially increase intracellular ROS levels. This increase in ROS is linked to Ras-induced mutagenesis which is one of the most common events in cancer (5). Sestrin 1 was also identified as a candidate gene for heterotaxia in humans (6). Recent studies have identified that AMPK is an essential mediator of tumor suppressor LKB1 and is suppressed in cancer cells (7). Since activity of AMPK is regulated by sestrin 1, this protein could be a promising target for cancer treatment and therapy. Sestrin 1 is approximately a 66.1kDa protein in humans (551 amino acids) and 55.4kDa protein in mouse and rat (462 amino acids).
The SESN1-selective antibodies were generated using peptide corresponding to rat SESN1 protein. SESN1-antibodies are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int.
