Sterol regulatory element binding protein 2
Sterol regulatory element binding-protein 1 (SREBP1/SREBF1) belongs to the SREBP family of transcription factors which are members of the basic helix-loop-helix leucine zipper (bHLH-Zip) transcription factor family (1). SREBP family of transcription factors play a vital role in regulating genes involved in biosynthesis and uptake. More specifically, they modulate the expression of enzymes implicated in the synthesis of cholesterol, fatty acids, triacylglycerol and phospholipids (2). These proteins are synthesized in their inactive form in the ER where they form complex with SREBP cleavage activating protein (SCAP). SCAP restricts SREBPs to the ER under high cholesterol concentrations. It is only under low concentrations that SREBP is transported to the golgi apparatus for activation (1). Here, SREBPs undergo two proteolytic cleavage steps to release the active transcription factor known as nuclear SREBP (nSREBP). Once activated, SREBPs can translocate to the nucleus and bind to sterol response elements (SRE) in the promoter/enhancer regions of target genes (1).
Similarly, the activated SREBF2 binds to the sterol regulatory element-1 (SRE1) (3). This is a key protein that regulates genes involved in cholesterol biosynthesis and uptake and it is required for lipid homeostasis (4). SREBF2 is also known to regulate LDL receptor gene (5). Animal studies have reported that mice livers that contained high levels of SREBF2 usually caused an increase in the rate of cholesterol and fatty acid synthesis in that animals when compared to mice whose livers contained normal levels of SREBF2 (6). SREBF2 is universally expressed in fetal and adult tissues. It is approximately a 136.9kDa protein (1141 amino acids).
The SREBF2 antibodies were generated using peptide corresponding to human SREBF2 protein. SREBF2 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int.
