The dominantly inherited two genes, BRCA1 and BRCA2 that contribute approximately 3-5% of breast cancer, account for approximately 85% of families with four or more cases of breast and ovarian cancer (1). Soon after these genes were discovered, it became apparent that specific founder mutations in these genes were present in different ethnic groups (2), at least 5 founder mutations were discovered that account for 75-85% of all BRCA1/2 mutations in specific ethnic group. However, for approximately one half of strongly hereditary breast cancer families, no BRCA1/2 mutation has been identified and other genes are believed to be important. Several known hereditary genes (BRCA1, BRCA2, CHEK2, TP53, ATM, FANCJ/BRIP1) are known to work in concert in DNA repair pathway which raises the possibility of other genes that might be involved in this pathway and are predispose to breast and other types of cancer (3). Recently identified gene Partner and localizer of BRCA2 (PALB2) is associated with breast cancer susceptibility in the Finland and other European countries. Finland population has shown mutations in PALB2 but these mutations are rare cause of breast cancer incidence in that country (4).
PALB2 is identified to play an important role in facilitating the function of BRCA2; nearly 50% of BRCA2 is associated with PALB2 and more than 50% of PALB2 forms a complex with BRCA2. Association of BRCA2 with PALB2 appears to be essential for BRCA2 anchorage to nuclear structures and for its function in homologous recombination and double-strand break repair (5). The PALB2 may also act by promoting stable association of BRCA2 with nuclear structures, allowing BRCA2 to escape the effect of proteosome-mediated degradation. Recent studies suggest that PALB2 is another form of FA gene also known ad FANCN, and FANCMN disease resembles FA arising from biallelic BRCA2 mutations in the affected children that are prone to embryonic tumors and early bone marrow failures. These patients exhibit increased chromosomal breakage after interstrand cross-linking agent exposure, exhibit growth retardation and various congenital malformations (6). The mutations in BRCA2 gene in prostate cancer have been associated with increased prostate cancer risk and account for around 2% of young onset of prostate cancer cases. Interactions of PALB2 with BRCA2 and its role in DNA double stranded break repair suggest a role of PALB2 in prostate cancer. Two previously unreported variants of PALB2, K18R and V925L were identified, neither of which is in a known PALB2 functional domain and both of which are unlikely to be pathogenic in prostate cancer (6). PALB2 (1186 aa) is highly conserved protein expressed in at least 2 variant isoforms in human and mouse/rats is localized on the short arm of chromosome 16p12.1 at position 23,521,983-23,560,127bp.
The PALB2-selective antibodies were generated against an epitope that is common in human and rat/mouse PALB2 protein near the C-terminal end of the protein.
