Survivin is a novel member of a group of proteins belonging to inhibitors of apoptosis (IAP). Survivin is expressed in cancer cells but is undetectable in normal, differentiated adult tissues. Survivin is expressed during embryonal development but lacks expression in terminally differentiated adult tissues. It has been shown that it is expressed in tumor cells from different cell lineages. It can inhibit apoptosis that is induced by a variety of factors, and plays important roles during mitosis and cytokinesis of cell. However, the mechanism of regulation of the survivin expression still remains unclear. Recent research suggest that surviving is implicated in regulation of cell division and control of apoptosis. In certain lung cancer cells production of nitric oxide down regulates the expression of surviving and induce apoptosis (1). Recently, several apoptosis inhibitors related genes have been found in various species, including human. These genes code for proteins that contain 1-3 Cys/His baculovirus IAP repeats plus a c-terminal RING finger. These structural motifs are thought to inhibit various steps in self induce apoptotic process. The overexpression of surviving is also implicated in resistance to chemotherapy of certain tumor cells. Thus, Survivin is associated with cytoprotectionand preservation of cell shape (2) , and its expressionin malignant cells may facilitate evasion from apoptosis andpromote aberrant mitotic progression, thus exacerbating aneuploidy.
Survivin is a 16 kDa protein with amino acids. The expression of Survivin in prenatal tissue is turned off and not found in non-neoplastic adult human and other species tissues. The expression of this important protein is turned on again in transformed cells and in the most common cancers of lung, colon, pancreas , breast, and proastate in vivo. The presence of survivin is believed to regulate and control the cell death and cell division by controlling a check point involved in cytokinesis and suppressing apoptosis. The surviving also inhibits Fas-mediated apoptosis in variouis immune cells (2). In lung cancinoma cells, surviving is a phosphorlyated by protein kinase B (3). One of the important step in survivin function is identified recently which require phosphorylation of survivin on Thr34 by the mitotic kinase p34cdc2-cyclin B1 (4) which inhibit p53 induced apoptosis (2). Phosphorylation of surviving by PKB has been shown to arrest the cells in G2/M phase (3). It was foundthat phosphorylation on Thr34 is required to maintain survivinexpression/stability in cancer cells and that ablation of p34cdc2 kinase in mitotically arrested cells results in loss of survivinlevels and dramatic enhancement of chemotherapy-induced anticanceractivity in vivo.
