Runt-related Transcription factor1 (Runx1) belongs to the family of Runt-related to a transcription factors comprising of 3 subtypes (Runx1-3).  Regulation of cell proliferation by RUNX proteins represents an evolutionarily conserved activity.  All vertebrates have three Runx genes and all three genes have P1 and P2 promotors. The Runx3 P1 and P2 promoter regions showed that a small (around 600 bp) P1 element could direct cell-type-specific expression with fidelity (1).   Runt-related transcription factor 3 (RUNX3) is a candidate tumor suppressor gene that is downregulated in various cancers that paly important rol es in cell proliferation and apoptosis. The mammalian AML/CBFalpha runt domain (RD) transcription factors regulate hematopoiesis and osteoblast differentiation. Like their Drosophila counterparts, most mammalian RD proteins terminate in a common pentapeptide, VWRPY, which serves to recruit the corepressor Groucho (Gro). The evolutionary conserved Runt domain harbors DNA binding and heterodimirizing activities.   The PEBP2/CBF is a heterodimeric transcription factor composed of alpha and beta subunits. The alpha subunits have PEBP2alfaA and PEBP2 alfaB and PEBP2 alfa C.  
the Runx3 gene codes a 51kDa protein, a heterodimier for Runx3 and a beta subunit forms a complex that binds to the core DNA sequence 5’-PYGPYGGT-3’ found in a number of enhancer and promotors.  The complex of Runx3:beta subunit can activate or suppress the transcription and also can inter actis with other transcription factors.  Runx3 is a tumor suppressor protein and  its gene is frequently transcriptionally silenced in different kind of cancer.  .  

Runx3- selective antibodies were generated against a peptide taken form N-terminal region spanning 200-250 amino acids. The 20mer peptide was conserved in Runx3 isoform 1 and isoform 2 of Runx3.  The antibodies do not cross react to other Runx subtypes. The anti-Runx1-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions.  The Runx1antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage.  FabGennix Int. Immunogen:                     The synthetic peptide taken from human Runx3 subtype 1 and 2 protein were taken form regioni corresponding to the aa acid sequences (KPF PDR FGD LER LRM RVT PS for Runx3 subtype ½) and (aa: asn sif dsf pty spt fir d from Runx3 isoform 1only).  The peptides wwere conserved in several other species.  The selected peptide was post-synthetically modified to achieve highest antigenicity before coupling to carrier protein using hetero bifunctional cross linker for immunogen preparation.

Conc.:          Affinity purified Runx3-301AP and Runx3-312AP antibodies were stabilized at a concentration 0.52-0.53mg/ml.

Appl.:        Runx3-301AP and Runx3-312AP antibodies were tested in ELISA and western blotting applications at 1:500 dilution using PC-Runx3 samples.  Antibody dilutions for these antibodies are for reference only, investigators are expected to determine the optimal conditions.  Application of this antibody in other protocols has not yet tested. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d.  Investigators using this antibody in protocols other than listed above can request a complimentary sample of this antibody. N.D not necessarily means the antibody is not suitable for that application, it simply means we have not yet characterized the antibody for that application.

Reactivity:  The antibody labels a strong band of Runx3of 51kDa in PC-Runx3samples and in several cell lines. The antibody also labels 2 faint bands of around 78 and 80kda, the identity of these bands is not known but may represent Runx3 splice variants.
