Chemokine (Chemoattractant Cytokines) are small peptides that are potent activators and chemo-attractants for leukocyte subpopulations and other non-hemopoietic cells.  Chemokine receptors (CXCR) belong to the super-family of G protein-coupled receptors (GPCR), which regulate the trafficking and activation of leukocytes, and operate as co-receptors in the entry of HIV-1 and proliferation and migration of immature neurons, glia and their precursors (1). Furthermore, chemokine receptors participate in the etiology and progression of various brain disorders, including AIDS dementia, neuro-inflammatory disease and neuroplasia, making them important potential therapeutic targets in these cases. Several subtypes of CXCR have been characterized (Table1).
Activation of naïve T cells by specific antigen and cytokines, up-regulate cell adhesion molecules (CAM) as well as chemokine receptors on their surface, which directs them to migrate towards the inflamed tissues. CXCR6, originally identified as a fusion cofactor for human immunodeficiency virus (HIV)-1 and simian immunodeficiency virus (SIV) and designated Bonzo, STRL33 and TYMSTR, is a member of the seven-transmembrane domain GPCR family. In humans, CXCR6 is preferentially expressed by subsets of memory/effector T cells, natural killer (NK) cells and NK T cells, and is also found on plasma cells. CXCR6 was present on 4–6% of CD4 and CD8 T cells in blood, normal lymph nodes (LNs) and the spleen, primarily on memory T cells (2). The only known ligand for CXCR6 is CXCL16 which is expressed as membrane bound form on antigen presenting cells such as dentritic cell, B cells and mono nuclear phagocytes, endothelial and smooth muscle cells. CXCR6 is also described as scavenger receptor for Phosphotidyl serine and oxidized lipoprotein on macrophages (3) however, this bifunctional role is not fully understood.
The CXCR6 receptor protein is approximately 42-45kDa (342 amino acids) protein which has 7 trans-membrane domains, characteristic of G-protein coupled receptors. The all CXCR sequence analyses suggest the presence of 7 trans-membrane domains, a characteristic feature of GPCR. The protein has putative N-glycosylation sites near the extra cellular N-terminal end of the proteins. The protein has a large 3rd intra cellular loop which interacts with G-proteins. The short carboxy terminal is intracellular and has putative post-translational sites. The Anti-CXCR6-selective antibodies were generated against conserved but unique sequences from human and rat/mouse specific CXCR6 protein, these sequences are not present on other CXCR or CCR receptors. The epitope specific rabbit polyclonal antibodies were affinity purified on an immobilized antigen based affinity chromatography. These epitope-specific antibodies strongly label CXCR6 protein in their respective western blot positive controls. Anti-CXCR6-selective antibodies are also available in the form of FITC-conjugated form for direct application in IHC and cell sorting assays, other fluorophore conjugation can be done upon request at a nominal charge.
