like IL 12, Interleukin 23 is composed of 2 subunits, P40 and P19, P19 is found to be distantly related to the p35 subunit found associated with IL12.  IL 23 is secreted in detectable quantities by Activated dendritic cells.  The IL 23 complex binds to IL12 receptor beta 1 subunit but failed to bind to IL12 beta 2.  Upon interaction with IL 12 receptor beta 1, IL 23 activates Stat 4 in PHA blast T cells.  IL-23 induces strong proliferation of mouse memory (CD4(+)CD45Rb(low)) T cells, a unique activity of IL-23 as IL-12 has no effect on this cell population. Similar to IL-12, human IL-23 stimulates IFN-gamma production and proliferation in PHA blast T cells, as well as in CD45RO (memory) T cells (2). Upon binding with IL23 receptor, this couples the JAK2.STAT4 pathway and induces the promotion of T cells towards the Th1 phenotype by strongly enhancing IFN-gamma Production.  There exist a significant sequence homology between cross species IL12P35 and IL23P19 subunits, in particular, the cysteins and the N-linked glycosylation sites which are conserved across the species.  Such sequence homology may be responsible for IL23 cross species biological activity (1).   There are 3 isoforms of IL23 receptors each is expressed in multiple splice variant form.  Human and mouse IL-23 exhibit some activities similar to IL-12, but differ in their capacities to stimulate particular populations of memory T cells. Thus, the ability of cells to respond to IL-23 or IL-12 correlates with expression of IL-23R or IL-12Rbeta2, respectively.

IL 23 P19 is a (189 aa) 19 kDa protein.  Experimental autoimmune encephalitis and multiple sclerosis is produced in mice lacking IL12 but not in mice targeted to disrupt the expression of IL23 or both IL 12 and 23, suggesting IL 23 expression in dendriatic cells may play a role in the pathogenesis of autoimmune diseases such as MS (3).  IL-23 induces the differentiation of naive CD4(+) T cells into highly pathogenic helper T cells (Th17/Th(IL-17)) that produce IL-17, IL-17F, IL-6, and TNF-alpha, but not IFN-gamma and IL-4 (4).  Keratinocytes induced cutaneous immune responses are also induced by IFN gamma inducing IL23, the IL23 expression was significantly higher in psoriatic lessioned skin.  

The IL23P19-selective antibodies were generated against unique antigenic sequences taken from human IL23P19 subunit. The 2 peptide antibodies are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Inc., will also provide limited quantities of antigenic blocking peptides for antibody IL23P19-101AP and IL23P19-112AP. Antibodies to other subunits of IL12 and IL23 and to other type of interleukins are also available from FabGennix International Inc. FabGennix Inc. will also conjugate antibodies with secondary enzymes (alk-Pase or HRP) or fluorescent probes upon request at a nominal cost.

Immunogen:            Synthetic peptides corresponding to positions: IL23P19 (aa 86-102) and for IL23P19-112AP (aa 162-179), sequences unique to IL23P19 protein and are conserved in many species.

Concentration:         IL23P19-101AP and IL23P19-112AP:    IgG concentration 0.65-1.25 mg/ml in antibody stabilization buffer.

Applications:           Antibody IL23P19-101AP and IL23P19-112AP are ideal for WB and ELISA applications, other applications have not been tested. These antibodies do not cross react to other interleukins or with other proteins. The species cross reactivity for these antibodies have not been fully examined. The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions for specific assay. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d.                                                 Reactivity:                This antibody detects a single band of approximately 19 in PC-IL23P19 samples.

Protocols:                Standard protocol for various applications (WB; IMM and IHC) of this antibody can be obtained by request, however, FabGennix Inc., strongly recommends investigators to optimize conditions for use of this antibody in their laboratories.

Form/Storage:          The antiserum is supplied in antibody stabilization buffer. The affinity-purified antibodies are isolated on immobilized antigen-affinity column and supplied as stabilized product. Store at -20oC for long-term storage. FabGennix Inc. does not recommend storage of very dilute antibody solutions unless they are prepared in specially formulated multi use antibody dilution buffer (Cat # DiluOBuffer). Working solutions of antibodies in DiluOBuffer should be filtered through 0.45ƒÝ filter after every use for long-term storage.
