he Nicotinic Acytylcholine receptor channel (nAChRs) that contain alfa7 gene products are the most abundant type of nicotinic acetylcholine receptors in nervous system. The alfa7 containing nAChRs have high affinity to Bungarotoxin toxicity and have calcium permeability compared to glutaminergic NMDA receptors. Physiologically, the a7-nAChRs modulated various functions including synaptic transmission, inflammation, and vasodilatation to apoptosis and are involved in learning and memory. The a7-nAChRs are also linked to Schizophrenia and are receptors for beta amyloid peptide (1).
Alfa7 nicotinic acetylcholine receptor forms homomeric pentamers when expressed in Xenopus oocytes and heteromeric pentamers with beta 2 subunits (2). There exist considerable differences in the pharmacology and biochemical characteristics of the alfa7nAchRs which indicate variability in the subunit structures. The widespread expression of beta 2 and alfa7 subunits in central nervous system suggest that these two subunits may form heteromeric pentamers in vivo. It is proposed that the heterogeneity in the alfa7-nAChRs might be caused by cell dependent expression of alfa7 subunit variants (3). In brain a novel alfa7-2 splice variant was detected that has a 87 base pairs (29 aa) insert that represent insertion of a novel exon (exon 4) between exon 4 and 5 of the alfa7-1 subunit. The alfa7-2 variant is expressed in CNS and periphery and when expressed in X oocytes it produces functional ACh-channels which resembles those of the slowly desensitizing a7-nAChRs.
The afla7-nAChRs variant-specific antibodies are produced using antigenic peptides from the exon 4 (29 amino acid) insert. The peptide was post-synthetically modified to achieve desired antigenicity before injecting into rabbits to make mono-epitope specific polyclonal antibodies. The antibody NACR-selective antibodies were affinity purified against immobilized antigen based affinity chromatography and are represented as pure IgG fractions stabilized in antibody stabilization buffer. The polyclonal antibodies strongly label a 58k Da alfa7-2 subunit in hippocampal membranes and in PC-NACR samples. The NCAR antibodies can be conjugated as HRP, alkaline phosphatase conjugates or with fluorophores (FITC, Rhodamine) for IHC, Confocal, WB analyses at a nominal price.
