Nineteen thousands genes are s[read over the 97-megabase genomic sequence of the nematode Caenorhabditis elegans. There exist significant predicted sequence homology between the 19,000 genes in C. elgans and other organisms and more than 40 percent of the predicted protein products are present in other organisms. There is a variety of repeated sequences, both local and dispersed. The distinctive distribution of some repeats and highly conserved genes provides evidence for a regional organization of the chromosomes. Several of the C. elegansproteins were surveyed for transcription factors and signaling genes families that have properties to regulate development in variety of other species, about half of the genes detect probable orthologs in other species, and about 10 to 25 percent of the genes are, at present, unique to C. elegans. Caenorhabditis elegans is also missing genes that are found in vertebrates and other invertebrates. Thus the genome sequence reveals universals in developmental control that are the legacy of metazoan complexity before the Cambrian explosion, as well as genes that have been more recently invented or lost in particular phylogenetic lineages (1).
The RNAi has been employed to study gene function in diverse helminthes, parasites, a dn nematodes. Double stranded RNA has been used to induce gene silencing in a variety of eukaryotes in several pathways. It is observed that RNA interference (RNAi) machiniery is present in ancestral eukaryotes and consist of one Argonute-like polypeptide, Piwi-like, dicer and RdRp proteins. In Nicotiana benthamiana, RdRPs have two closely related genes, one is similar to SDE1/SGS2 and the other,a sialic acid inducible RdRP1m both are 90% identical in sequence (2). The RdRP3 gene is locatd on chromosome II in C. elegans and has 1780 aa (~215 kDa).
RdRP3-selective antibodies were generated using cyclic peptide methodology, the peptide was selected form the RdRP1sequence of C. elegans from position 11776-1780. The RdRP3-selective antibodies were affinity purified against immobilized antigen based affinity chromatography and are represented as pure IgG fractions stabilized in antibody stabilization buffer. The polyclonal antibodies strongly label a ~225kDa protein form PC-RdRP3 sample on Western blots. The RdRP3 antibodies are also available in FITC conjugates and FabGennix will conjugate this antibody with other fluorophores like rodamine, biotin, avidin and enzymes at a nominal charge. Immunogen: Synthetic peptide from C. elegans RdRP3 (Rrf3) protein corresponding to the terminal 15 amino acids. Th peptide was covalently modified to achieve desired antigenicity and coupled to a carrier protein.
Concentration: RdRP3-101AP = 0.63-0.65mg/ml of antibody in stabilization buffer
Reactivity: The antibody labels a diffuse broad band of 210-225kDa in PC-RdRP3 and in tissue extracts of C. elegans.
Applications: Antibody RdRP3-101AP is ideal for ELISA and Western blotting, its use in other applications have not been established and tested. The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions for specific assay in his/her laboratory. Other applications have not been tested. Suggested Dilutions: WB: > 1:500; Imm & i.p pull-down assays: n.d. N.D. does not necessarily mean antibody is not suitable for application, investigators using this antibody in applications not listed here can ask for a complimentary sample of this antiobdy.
