Glutamate Receptor 2
Ionotropic Glutamate Receptor 2 (GluR2) is a predominant excitatory neurotransmitter in mammalian brain located at human chromosome 4q32-33 (1). It belongs to a family of alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA) receptors (2). GluR2 plays a crucial role in controlling the calcium permeability of AMPA receptors and therefore, its presence or absence greatly influences AMPA receptor properties and synaptic functions. GluR2 mRNA ordinarily undergoes post-transcriptional editing which is essential for brain function. AMPA receptors containing edited GluR2 have low conductance, low calcium permeability and confer a linear current-voltage (I-V) relationship. AMPA receptors lacking an edited GluR2 subunit have high calcium permeability and also exhibit inward rectifying I-V relationship (3). Defective GluR2 RNA editing might also play a role in the development of amyotrophic lateral sclerosis (ALS) (4).
GluR2 is among the most common and abundant receptor in striatum and projection neurons. In rats, all striatal projection neurons express GluR2 (5). It is highly concentrated on the postsynaptic neuronal membranes of the superficial dorsal horn and its removal might contribute to spinal central sensitization in persistent inflammatory pain (6). It is expressed in cortex, hippocampus and cerebellum. It is approximately a 106kDa protein (886 amino acids).
The GluR2-selective antibodies were generated against purified Glutamate receptor 2 (GluR2) protein. GluR2-antibodies are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Inc. will also provide limited quantities of antigenic blocking protein for GluR2 -201AP antibody. Antibodies to GluR2 (GluR2 -201AP) will label ~106kDa protein in Western blot positive control samples for GluR2 and several other tissues. FabGennix Inc. will also conjugate antibodies with secondary enzymes (alk-Pase or HRP) or fluorescent probes upon request at nominal cost.
