Fam38A (Piezo1)

Mechanical stimuli are essential for many physiological process including touch, pain sensation and blood pressure regulation. Mechanically activated cation chanel activities are attributed to two proteins with multi-trans-membrane spanning domains proteins Fam38A and Fam38B also known as Piezo 1 and Piezo 2 (1).  The Fam38A/B are expressed in invertebrates, plants and protozoa.  Fam38A/B are expressed in several tissues, and over expression of Fam38A/B induced kinetically distinct mechanical currents. 

The cell surface receptors of integrin family of heterodimeric proteins are essential to cell-cell matrix adhesion.  Any changes in the integrin binding affinity or its gene expression are central to a variety of disease progression including inflammation, cardiovascular disorders and cancer.  Fam38A, a multi-trans-membrane domain protein , mediates integrin activation by recruiting small GTPase R-ras to endoplasmic reticulum, which activates Ca-activated protease calpain by increasing Ca-release from cytoplasmic stores. The Fam38A-induced activation of integrin is blocked by inhibition of R-ras or calpain activity (2).  Beta-amyloid (beta) deposition and senile plaque-associated astrocytes are common neuropathological observations in Alzheimer’s disease (AD).  The association of Abeta with astrocytes, the predominant cell types in brain, significantly influences the exacerbation of AD. A novel Abeta induced gene, Mib (human counter parts covers KIAA0233) has been identified in AD patients astrocytes.  The Mib transfected C6 cells express Mib protein in the endoplasmic reticulum.  In AD brain samples, the Mib protein is expressed in neurons and quiescent astrocytes while in non-AD brains the expression is limited to neurons.   

The Fam38A-selective antibodies were generated against unique antigenic sequence between extracytoplasmic domain between TMD14 and TMD15 region of the Fam38A protein.  The antibodies to Fam38A are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer.  FabGennix Int. Inc., will also provide limited quantities of antigenic blocking peptides for Fam38A-101AP.  Antibodies to Fam38A (Fam38A-101AP) will label ~210kDa diffuse bands in PC-Fam38A samples.  FabGennix Int. Immunogen:            Synthetic peptides taken form between TMD14 and TMD15 of the Fam38A gene, peptide sequence is unique to Fam38A and was not present in Fam38B protein and is present in a wide range of species.   

Concentration:         Fam38A-101AP:  IgG concentration 0.66-0.62 mg/ml in antibody stabilization buffer.

Applications:           Antibody fm38A-101AP is ideal for WB and ELISA applications, other applications have not been tested.  This antibody does not cross react to other ion channel proteins.  The species cross reactivity for this antibody is not examined.  The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions for specific assay.  WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d.  ELISA <1:10,000 

Reactivity:               This antibody detects multiple (3-4 bands) bands in the 250-200kDa molecular weight range probably belongs to multiple splice variants as all of them were blocked by excess antigenic blocking peptide.  The antibody does not cross reacts with other proteins of the ion channel proteins.   

Protocols:                Standard protocol for various applications (WB; IMM and IHC) of this antibody can be requested by calling Technical support line, basic information on this antibody is provided with the product specification sheet, however, FabGennix Inc.  strongly recommends investigators to optimize conditions for use of this antibody in their laboratories.  

Form/Storage:         The affinity-purified antibodies are supplied as stabilized product.  Store at -20^oC for long-term storage.  FabGennix Inc. does not recommend storage of very dilute antibody solutions unless they are prepared in specially formulated multi use antibody dilution buffer (Cat # DiluOBuffer).  Working solutions of antibodies in DiluOBuffer should be filtered through 0.45ƒÝ filter after every use for long-term storage.
