Solute carrier family 6, member 3 which is commonly known as sodium-dependent dopamine transporter (DAT1) is a plasma membrane transport protein located on human chromosome 15p15.3 (1). It belongs to the sodium and chloride-dependent neurotransmitter transporter family and plays a key role in controlling dopamine-mediated neurotransmission (2). The 3’ untranslated region of this gene contains a 40bp variable number of tandem repeats, which can be present in 3-11 copies. Variations in the number of repeats of this VNTR have been implicated in various disorders including attention deficit hyperactivity disorder (ADHD), alcohol and nicotine addiction, parkinson’s disease (PD) and idiopathic epilepsy (3). The allele with 10 copies of this sequence has been implicated in dopamine transporter density in mammals (4). Studies have found abnormal densities of dopamine transporter in the brain region of patients with ADHD. Stimulant medications such as amphetamine and methylphenidate which are efficient in reducing ADHD symptoms inhibit the dopamine transporter and dopamine uptake and treatment with these medications decreases the densities of DAT1 in the brain region. Therefore, DAT1 is a promising target in treatment of ADHD (5).
The 3’ untranslated region of DAT1 is also associated with cold pain tolerance. This gene was shown to interact with early drug exposure (heavy or regular) of the maturing adolescent brain to predict substance use or abuse in young adulthood; therefore, it is also associated with alcohol and nicotine consumption (6). Recently it was reported that the promoter region of DAT1 is implicated in the etiology of schizophrenia and the immunoreactivity of this gene is significantly reduced in peripheral blood lymphocytes in parkinson’s disease (7). DAT1 is expressed in the brain and it is approximately a 74.4kDa protein (620 amino acids).
The SLC6A3 antibodies were generated using peptide corresponding to human SLC6A3 protein. SLC6A3 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int.
