The cellular levels of Cyclic GMP (cGMP), a key messenger in several signal transduction pathways, are maintained by the orchestrated activities of two opposing enzymes: synthesizing gualylyl cyclases (GC) and hydrolyzing phosphodiesterases (PDEs). The synthesizing enzymes (GCs) are found in two forms: cytosolic (soluble) and membrane-bound (particulate). In last several years new members of particulate guanylate cyclase family have been identified and significant progress is made in understanding the molecular mechanisms that underlie the regulation of these family of enzymes. In response to G-protein couples receptor stimulation, the cGMP can be produced from GTP by either cytoplasmic, soluble guanylate cyclase (sGC) are heterodimers (a and b polypeptide chains), that are stimulated by nitric oxide and carbon monoxide or by particulate membrane-bound guanylyl cyclases which are activated by a complex mechanism by natriuretic peptides.  Particulate GC (PGCs) have 7 different isoforms, PGC-A through PGC-G and are expressed in most tissues in isoform specific manner (See Table 1).  There is significant structural homology among various PGCs, there is a large N-terminal extracellular domain (ECD), a single TMD and a large intracellular domain with protein kinase activity (KLD), a C-terminal catalytic domain (CD) and in between is a dimmerization domain (DD). It has been shown that a significant number hippocampal astrocytes (67%) contained both soluble and particulate guanylate cyclases in the same cell (2).  
The Anti-sGCA1-selective antibodies were generated against conserved sequence near the carboxy terminal end of the protein (aa 685-704) that are unique to sGC-Alfa1 subunit and is conserved in human with a single conservative substitution (Asp to Glu) in human protein. Soluble guanylate cyclase alfa1 is a 704 amino acid long protein. The sGCA1-selective antibodies are affinity purified against immobilized antigen based affinity chromatography which yielded epitope -pecific antibodies.  The sGCA1 antibodies label a 84-86kDa protein in soluble guanylate cyclase A1 Western blot positive control. Anti-sGCA1-selective antibodies are also available in affinity-purified form for confocal, WB and IHC analyses. FabGennix Inc. will also conjugate antibodies with fluorescent probes upon request at extra charge.
