Septin 9

Septin 9 (Sept9) belongs to the septin family of GTPases. GTPases are central regulators in signal transduction pathways in every aspect of cell biology (1). Septins were first discovered in budding yeast. Septins bind to and hydrolyze GTP and most of these proteins polymerize to form filaments which may form cytoskeletal system that organizes higher-order structures and where membrane movement occurs such as cytokinesisand secretion (2). Septins were first discovered in yeast Saccharomcyes Cerevisiae, they are also found in animals and fungi but not in protozoa or plants (3). Septins are implicated in cytokinesis, exocytosis, tumorigenesis, apoptosis, synaptogenesis, neurodegeneration, and in functions involving changes in cell shape and in the specialized regions of the cell cortex (4). Humans have twelve septin genes, some of which generate multiple polypeptides either by alternative splicing or alternative translation start sites (2). The function of individual protein in septin family usually depends on its assembly into oligomeric complexes and highly ordered polymers (5).
Septin 9 is the largest transcript of the Spetin family, Septin 9 is implicate din the tumorigenic transformation of mammary epithelial cells. Up-regulation of Septin 9 stabilizes JNK by delaying its degradation (6). Alterations in human septin-9 (SEPT9) levels have been linked to multiple cancers, whereas mutations in SEPT9 cause the episodic neuropathy, hereditary neuralgic amyotrophy (HNA). The Septin 9 is a potential biomarker for colorectal cancer, patients with colorectal cancer exhibit have high levels of methylated DNA primarily due to increased activity of Septin 9 (6). Septin 9 is located on chromosome 17q25.3. It is expressed in brain, heart, platelets and other tissues with high expression in aorta. Septin 9 is approximately a 69kDa protein (568 amino acids).
The Sept9 antibodies were generated using peptide corresponding to human Sept9 protein. Sept9 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Immunogen: Synthetic peptide unique to Sept9 with in the amino acids 1-60 was covalently modified to get the desired antigenicity before conjugation to a carrier protein.
Concentration: Sept-901AP and Sept-FITC: IgG concentration 0.65-0.69mg/ml in antibody stabilization buffer.
Applications: Antibody Sept-901AP is ideal for WB and ELISA applications, other applications have not been tested. The species cross reactivity for this antibody has not been fully elucidated. The dilution for this antibody is for reference purposes only, investigators are expected to determine the optimal conditions for a specific assay. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d. ELISA <1:10,000. Application of this antibody in protocols not listed here does not necessarily exclude its use in such procedures. Reactivity: This antibody detects a band of approximately 69kDa in PC-Sept9 samples.
