Eukaryotic protein synthesis initiation factor
The capped-mRNA binding protein, eukaryotic protein synthesis Initiation factor 4E (eIF-4E) is directly involved in the initiation of protein synthesis in eukaryotic cells. It has been shown that eIF-4E phosphorylation at serines by insulin dependent mechanism coincides with increased protein synthesis in various tissues (1-3). Numerous other extra cellular stimuli also phosphorylate eIF-4E via protein kinase C and insulin-stimulated protamine kinases (cPK). Conversely, dephosphorylation of eIF-4E results in decrease protein synthesis rate. It has been shown that cPK and PKC phosphorylates eIF-4E at Serine209 and threonine210. The phosphorylation of serine 209 activates the eIF-4E protein synthesis activity, protein phosphatase 2A (PP2A), a major cytoplasmic serine-, threonine-phosphatase preferentially dephosphorylates insulin-dependent phosphorylated eIF-4E factor at threonine 210.
The eIF-4E selective antibodies were generated against purified human recombinant eIF-4E protein. The antibodies eIF-4E are affinity purified over immobilized antigen based chromatography. The purified antibodies are stabilized in antibody stabilization buffer containing preservatives. FabGennix Int.
