Nucleophosmin (NPM/B23) is a multifunctional oncoprotein whose protein expression levels dictate cellular growth and proliferation rates.  The 3’ untranslated region (UTR) of the NPM messenger RNA is sufficient to modulate its translation by mTOR signaling mainly due to binding of far upstgream element (FUSE) binding protein (FUBP1). Over expression of FUBP1resulted in to translational repression of NPM mRNA , whereas depletion of FUBP1 caused a dramatic increase in NPM expression and resulted in to enhanced cell proliferation (1).  FUBP1 is a key regulator of the cell proliferation and growth via its ability to selectively bind the NPM 3’ UTR region. Human c-myc expression is also under regulation  by an activator FUBP1 and a repressor, the FUBP1-intercting repressor FIR (2). Under physiological condition, FIR is always in monomeric form but upon binding with FUBP1 it forms a complex FIR2-FUBP1 mediated by RRMs (3). 

The FUBP1 and CIC (drosophila gene capicua) genes have shown mutation resulted in formation of truncated from of CIC and FUBP1 in oligodendroglioma tumors.  These results suggest FUBP1 and CIC genes may play critical role in pathology of oligodenroglioma (3).  The FUBP1 gene is located on chromosome 19q .  At least three Far upstream-element binding proteins (FUBP1-3) have been characterized to bind specifically to TG microsatellite in the untranslated 3’UTR region.  The protein encoded by FUBP1 binds to single stranded DNA, in particular the far-upstream element (FUSE) of MYC, a well-studied oncogene. 

MUBP1- selective antibodies were generated against peptides form unique region on FUBP1 protein corresponding to amino acid 626-642 near the C-terminal end of protein. This region was not only antigenic but also lies on the surface of the protein in native or in partially denatured FUBP1 confirmation.  The anti-FUBP1-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions.  The FUBP1antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage.  FabGennix Int. Immunogen:                     Synthetic peptide taken from human FUBP1 protein taken from within the C-terminal domain of FUBP1 corresponding to amino acid sequence: qtspqgpmqhppapc. The peptide was conserved in several other species.  The selected peptide was post-synthetically modified to achieve highest antigenicity before coupling to carrier protein using hetero bifunctional cross linker for immunogen preparation.                 

Concentration: Affinity purified FUBP1-101AP and FUBP1-FITC antibodies were stabilized at a concentration 0.53-0.55mg/ml for long-term storage.

Applications:   FUBP1-101AP antibody was tested in ELISA and western blotting applications at 1:500 dilution using PC-FUBP1 samples.  Antibody dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions.  Application of this antibody in other protocols has not yet tested. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d.  Investigators using this antibody in protocols other than listed above can request a complimentary sample of this antibody. N.D not necessarily means the antibody is not suitable for that application, it simply means we have not yet characterized the antibody for that application.

Reactivity:  The antibody labels a strong diffuse band of FUBP1of 80kDa in PC-FUBP1 samples and in several cell lines. The antibody also labels bands of 60 and 42 kD, the identity of these band is not known but may represent FUBP1 splice variants.
