The peptidyl prolyl cis/trans isomerase Ess1/Pin1 is essential for mitosis progression in yeast cells and is hypothesized to perform the same role in mammalian cells. PIN1-deficinet mice though underwent normal development, the embryonic PIN1 lacking fibroblast exhibited significant deficiency in their ability to restart proliferation in response to serum stimulation after a G(0) arrest suggesting that PIN1 is required for cell cycle progression from G(0) arrest and in mitosis progression in normal mammalian cells (1). Pin 1interacts with SIL, an early transcription factor essential for embryonic development in T-cell leukemia-associated translocations and hyper phosphorylate it in a cell cycle–dependent manner (2).  By virtue of its activity, PIN1 presumably interacts with NIMA and attenuate its mitosis-promoting activity. The PIN1 displays a preference for a acidic residue N-terminal to the isomerization of proline bond when proline is flanked by serine and threonine catalyzing p-Ser/P-Thr-Prol cis/trans isomerization. The interaction of PIN1 with tau protein in brain implicated its protective in neurodegenerative diseases such as Alzhiemers and frontotemporal dementia with Parkinsonism linked to chromosome 17. PIN1 binds and isomerizes tau and has a protective effect in tau stability (3). 
PIN1 also functions as a positive regulator of neuronal death through a c-Jun-dependent mechanism. The ectopic PIN1 promoted caspase-dependent apoptosis of neurotrophic growth factor (NGF) maintained neurons. The PIN1 knockout condition accumulated phosphorylated c-Jun, inhibited the release of cytochrome c and delayed the cell death suggest a novel mechanism by which PIN1 promotes apoptosis involving activation of c-Jun (4).  PIN1 also interacts with proto oncogene c-Myb thereby increasing the c-Myb trans-activation activity providing a new paradigm by which post-translational modifications phosphorylation controls c-Myb activity (5).
