Spingosine Kinase 1 (Sphk1) is a key lipid enzyme of the sphingolipid metabolic pathway and it catalyzes the phosphorylation of sphingosine, converting sphingosine into pro-growth and anti-apoptotic sphingosine 1-phosphate (S1P) (1). S1P plays a major regulatory role in tumor cell growth, survival, invasion, and angiogenesis (2). Sphk1, while producing pro-growth S1P, also decrease intracellular levels of pro-apoptotic ceramide. By regulating intracellular balance between S1P and its metabolic precursors, ceramide and sphingosine, Sphk1 plays a key role in determining whether a cell proliferates or undergoes apoptosis or growth arrest (3). Sphk1 also acts as a key regulator of acute and delayed monocyte activation. Its levels were significantly upregulated in CXC chemokine ligand 4 (CXCL4)-treated monocytes (4). Furthermore, inhibition of Sphk1 resulted in the reduction in monocyte cell survival, cytokine expression, and release of oxygen radicals.
The upregulation of Sphk1 mRNA and its expression was reported to be significantly higher in various tumors. Therefore, Sphk1 may also act as an oncogene (5). Sphk1 was inhibited in prostrate cancer cells and animal models undergoing anti-cancer treatments (6). Sphk1 levels were also up-regulated in stimulated phagocytes of human and mice with severe sepsis. Inhibition of this protein inhibited phagocyte production of endotoxin-induced pro-inflammatory cytokines that contribute to sepsis and also protected mice against sepsis (7). Sphk1 is located on human chromosome 17q25.2. It is highly expressed in adult liver, kidney, heart and skeletal muscle. In rat and mouse, Sphk1 is approximately a 45.9kDa protein (383 amino acids). In humans, it is a 56.4kDa protein (470 amino acids).
The Sphk1 antibodies were generated using peptide corresponding to human Sphk1 protein. Sphk1 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int.
