Septin 1 (Sept1) belongs to the septin family of GTPases. GTPases are central regulators in signal transduction pathways in every aspect of cell biology (1). Septins bind to and hydrolyze GTP and most of these proteins polymerize to form filaments which may form cytoskeletal system that organizes higher-order structures (2). Septins were first discovered in yeast Saccharomcyes Cerevisiae, they are also found in animals and fungi but not in protozoa or plants (3). Septins are implicated in cytokinesis, exocytosis, tumorigenesis, apoptosis, synaptogenesis, neurodegeneration, and in functions involving changes in cell shape and in the specialized regions of the cell cortex (4). Humans have twelve septin genes, some of which generate multiple polypeptides either by alternative splicing or alternative translation start sites (2). The function of individual protein in septin family usually depends on its assembly into oligomeric complexes and highly ordered polymers (5). Septin 1 is also involved in ciliary movements in microorganism.
Septin 1 may play a role in chromosome segregation. It is phosphorylated by Aurora-B, an important serine/threonine kinase required for chromosome segregation and cytokinesis, and these two proteins partially colocalize to the mid-body during cytokinesis (6). Sept1 may also contribute to the formation of neurofibillary tangles in Alzheimers disease and the expression of this protein has been linked with development of cancer. Sept1 was found to be overexpressed in oral squamous-cell carcinoma (7). Sept 1 is located on human chromosome 16p11.1. It is highly expressed in lymphoid and haematopoietic tissue. It is approximately a 46kDa protein (372 amino acids).
The Sept1 antibodies were generated using peptide corresponding to human Sept1 protein. Sept1 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int.
