Collapsing mediated response proteins (CRMPs) form homo- and hetero tetramers and facilitate neuron guidance, growth and polarity. It is abundantly expressed in brain and regulate neurite outgrowth.  The CRMP family consists of 5 subtypes (CRMP1-5) and are also known as dihydroxypyrimindase 1-5 (DRP1-5).  The CRMPs promotes microtubule assembly and is required for Sema3A-mediated growth cone collapse and also plays a role in synaptic signaling through interactions with calcium channels.  CRMP2 protein regulates endocytosis where two proteins MICAL-L1 and EHD1 are important regulatory proteins that control key endocytotic transport steps. CRMP2 interacts with MICAL-L1 in non-neuronal cells (1).  CRMP2 interacts with tubulin dimers and kinesin and negatively regulates s dynein-based transport in neuronal cells (2). 

The mammalian target of rapamycin (mTOR)is an important regulator of neuronal development and function. mTOR inhibitor, rapamycin regulates neuronal polarity by suppressing the expression of Tau and CRMP2 via mTOR effector 70-kDa ribosomal protein S6 kinase (p70S6K)p70 (2). It is suggested that abnormal activation of CDK5 is associated with neurodegeneratative disorders and it disrupts mature neuronal circuitries and neurogenesis. CDK5 and other kinases hyper-phosphorylate down-stream targets of these kinases in brain such as doublecortin, nestin, and CRMP2 (2). This gene has been implicated in multiple neurological disorders, and hyperphosphorylation of the encoded protein may play a key role in the development of Alzheimer’s disease. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene.

CRMP2-selective antibodies were generated against peptides form unique region near N-terminus of CRMP2 protein with in aa 1-100.  The epitope is conserved in several other species beside human but was unique to CRMP2.  The CRMP2 antibodies are affinity purified over immobilized antigen based affinity chromatography and fully characterized for their listed applications and cross reactivity.  The CRMP2 antibodies are also available as FITC-conjugates, other types conjugates can be ordered at a nominal charge. Immunogen:      Synthetic peptides corresponding to amino acids (qrd vah lgp dpq ppy srq grr agg ep) form CRMP2 protein. The peptide was covalently modified to achieve desired antigenicity before coupling to carrier protein.

Concentration:  CRMP2-201AP and FITC-CRMP2                IgG concentration 0.55-0.65 mg/ml in antibody stabilization buffer. 

Applications:      Antibodies CRMP2-201AP is ideal for WB, its application in IHC, IF, confocal or immunoprecipitations protocols have not yet tested and standardized. The dilutions for this antibody is for reference only, investigators are expected to determine optimal conditions for specific assay.  It is highly recommended that user optimize conditions for specific use of this antibody in a particular application with positive and negative controls.   WB: 1:450; IMM & i.p pull-down assays:>n.d.  IHC n.d.  Application of this antibody in protocols not listed here does not necessarily exclude its use in such procedures. Reactivity:           This antibody labels two major bands in PC-CRMP2 of approximate MW of 83kDa and a smaller band of 35kDa, the identity of smaller protein is not known but could represent a proteolysis product.
