RhoGAP_SYD1: RhoGAP (GTPase-activator protein [GAP] for Rho-like small GTPases) domain present in SYD-1_like proteins. Syd-1, first identified and best studied in C.elegans, has been shown to play an important role in neuronal development by specifying axonal properties. Fast chemical synaptic transmission is mediated by precisely regulated neurotransmitter release from synaptic vesicles at specialized presynaptic sites. This compartment called the active zone and comprises a unique set of proteins. The active zone is organized by proper orientation of Syd-2/Liprinalfa, Syd-2, ELKS/ERC/CAST, Regulator f synaptogenesis 1 (RAS1) and serine threonine kinase SAD-1. The synapse defective 1, syd-1) is a multi-domain Rho GAP-like protein that is required for the proper assembly of active zones in synapse (1). Syd-1 specifically localizes to to discrete compartment of the active zone edge. Mutation in Syd-1led to impaired locomotion and reduced life span in fruit flies that is rescued by expression of Syd-1 in nervous system (2).
Syd-1 is a Probable GTPase activator for the Rho-type GTPases by converting them to an inactive GDP-bound state. Regulates the localization and assembly of presynaptic components during presynaptic development and is required for specifying the identity of axons during initial polarity acquisition. In these roles it is thought to act cell autonomously downstream of syd-1 and syd-2 and upstream of syd-2, possibly as a positive regulator of the Syd-2. This Syd-1 is required for proper functioning of Syd-2. The Syd-1 is expressed in the nerve ring of embryos, and in the ventral chord motor neurons. IN adults, the expression is seen along the dorsal and ventral nerve cords. The protein contains several functional domains including a C1 domain, PDZ/DHR domain and a Pho-GAP domain. Syd-1 exhibit polymorphism in prostate cancer cells and Syd-1 is phosphorylated at S231, 235, 244, 247 and 683 in cervix carcinoma. Human Syd-1is 88 kDa (737aa) protein highly conserved in many species.
The Syd-1 antibodies were generated using peptide corresponding to human Syd-1 protein. Syd-1 antibodies are affinity purified over immobilized antigen based affinity chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer. FabGennix Int. Immunogen: Synthetic peptide corresponding to unique epitope on Syd-1. The C-terminal peptide sequence was selected from within amino acids spanning 660-735. These peptide was covalently modified post-synthetically to achieve desired antigenicity before coupling to a carrier protein.
Concentration: Syde-101AP and Syde-FITC IgG concentration 0.68-0.67 mg/ml in antibody stabilization buffer.
Applications: Antibody Syde-101AP is ideal for WB and ELISA applications, other applications have not been tested. The species cross reactivity for this antibody is based on epitope homology and has not yet experimentally confirmed. The dilutions for this antibody is for reference only, investigators are expected to determine the optimal conditions for specific assay. WB: > 1:500; IMM & i.p pull-down assays: n.d; IHC n.d. ELISA <1:10,000. Reactivity: This antibody detects two bands of approximately 93 and 89kDa in PC-Syde samples. the identity of 94kDa band is not known but may represent phosphorylated form of Syde. In tissue extracts antibody also labels few smaller bands the identity of these bands is not known but may represent proteolytic degradation products.
