Sirtuins are constitutively expressed in all domains of life. It was first identified in Saccharamyces cerevisiae as sirtuin, Sir2 (silent information regulator 2) which regulates ribosomal DNA recombination, gene silencing, DNA repair, chromosomal stability and longevity. The activities of epigenetic regulators such as histone acetyltransferases (HDACs), methyltransferases, chromatin-remodeling enzymes, etc., play a fundamental role in the control of gene expression by covalently modifying the local state of chromatin. The science of epigenetic regulation is a fairly new in gene regulation and not much is known about the regulation of these epigenetic-regulators by alternative splicing. The chromatin modulations is achieved by a series of chromatin modifying enzymes by either introducing histone modifications or by inducing ATP-dependent nucleosome remodeling. The regulatory state of a gene is explained by extend of covalent modifications (acetylation, methylation and ATP-dependent phosphorylation of histones as explained by “histone code hypothesis”. 
There are several mammalian homolog of yeast protein Sirtuin 2 (Sir 2) including Sirtuin 3, Sirtuin 4 and Sirtuin 5. The yeast homologs are involved in extension of yeast longevity by caloric restriction. The localization of sirtuin 3, 4 and 5 are localized in distinct compartment of mitochondrion. When Sir 3 and 5 are coexpressed the localizxation of Sir 3 changed from mitochondria to nucleus (1). The cytoplasmic sirtuin 2 is the most abundant sirtuin family member expressed in adipocytes, its expression is down regulated in 3T3 cells during their transformation in to adipocytes, the process of morphological differentiation is inhibited by over-expression of sirtuin 2 (2).  In 3T3 cells the sirtuin 2 controls the acetylation of Fox 1 protein (1). The adiopocyte differentiation is modulated by sirtuin 2 via acetylation of Fox1 protein. The microtubule regulatory alpha-tublin deactylases, histone deacetylase-6 (HDAC6) and Sirtuin 2 are the major players in cellular development of nervous system where all major cell types undergo cellular differentiation, generate unsurpassed quantities of plasma membranes and extend cellular processes to communicate and regulate the activities of other cells. The HDAC6 is abundantly expressed in Purkinje cells in hippocampus and Sirtuin 2 is mainly associated with myelin sheaths. The expression of these proteins in post-mitotic oligodendrocytes suggests their novel function in process out-growth and membrane remodeling by Sir2 in conjunction with the microtubule destabilizing protein Stathmin 1 during the early myelognesis. The lack of Sir 2 in oligodandrocytes of spastic paraplegic mouse model deficient in proteolipid protein (PLP)/DM20 suggest that lack of Sir 2 expression is post-translational and is mainly due to absence of transport carrier P:P/DM20 (3). 
The Anti-SIR3-selective antibodies are generated against unique sequence (aa 203-221 mouse Sirtuin 3 sequence) that is unique to SIR3 protein and is common in human, mouse , cat, bovine and several other species.
